GLP1R&CTR/RAMP3 Dual Reporter Cell
| I. Background | |
| 胰淀素（Amylin）受體的三種類型的已被確定，即 AMY1，AMY2 和 AMY3。胰淀素 受體屬于 B 類 GPCR，是異二聚體復合物，由降鈣素受體（CTR）與受體活性修飾蛋白 （RAMPs）結(jié)合形成。該受體的 CTR 組分具有相同的 GPCR 結(jié)構(gòu)：7 個跨膜相互連接的α- 螺旋，將細胞外信號傳遞到細胞內(nèi)細胞質(zhì)信號級聯(lián)。RAMP 有三種類型（RAMP1、 RAMP2 和 RAMP3），它們共享 31%的氨基酸序列，具有相似的基本結(jié)構(gòu)。RAMP 是將 CTR 轉(zhuǎn)運到細胞膜并調(diào)節(jié)其配體特異性的重要肽，它可以改變特定 GPCR 的藥理學、功能 和細胞運輸。 GLP-1R 的內(nèi)源性配體是 GLP-1（7-36）和 GLP-1（7-37），它們是胰高血糖素原翻譯 后加工的產(chǎn)物。GLP-1R 主要通過 Gαs/cAMP 通路發(fā)出信號，直接導致葡萄糖誘導的胰島 素顆粒分泌；但有證據(jù)表明 GLP-1R 可與 Gαq 和其他 G 蛋白偶聯(lián)；GLP -1R 激活后，C 末 端發(fā)生磷酸化，進一步募集β-抑制蛋白，導致受體內(nèi)化和脫敏。 GLP1R 與 AMY3 的聯(lián)合作用是當前代謝性疾病研究的前沿方向。結(jié)合 GLP-1 受體激 動劑和胰淀素受體激動劑被認為具有協(xié)同減肥和降糖效果。 | |
