ANG/TIE2 Effector Reporter Cell
| I. Background | |
| ANG（血管生成素）是調控血管穩(wěn)定的關鍵分泌蛋白，包含 ANG1 和 ANG2 兩種核心 亞型。ANG1 是由周細胞分泌，通過激活 TIE2 受體維持血管完整性與成熟度。ANG2 是由 應激內皮細胞自分泌，作為血管失穩(wěn)開關，低 VEGF 環(huán)境誘導血管退化（如糖尿病視網膜 病變），而高 VEGF 時則促進病理性血管新生（如腫瘤血管生成），其表達受缺氧（HIF- 1α）和炎癥因子（TNF-α）顯著誘導。 TIE2（激酶受體）主要表達于血管內皮細胞表面的受體，胞外區(qū)含免疫球蛋白 域 （ 結 合 ANG ） 、 EGF 樣 域 （ 介 導 二 聚 化 ） ； 胞 內區(qū)含激酶活性位 點 （Y1100/Y1108）。被配體激活后通過 PI3K/AKT 和 MAPK 通路調控內皮細胞存活、遷移 及血管屏障功能。在病理狀態(tài)下（如膿毒癥、腫瘤），TIE2 信號異常導致血管滲漏或異常增生。 | |
