BAFFR Effector Reporter Cell
| I. Background | |
| BAFF（B 細(xì)胞激活因子，B Cell Activating Factor），也稱為 TNFSF13B（Tumor Necrosis Factor Superfamily Member 13B）或 BLyS（B Lymphocyte Stimulator），是一種屬 于腫瘤壞死因子（TNF）超家族的細(xì)胞因子。BAFF 主要由單核細(xì)胞、巨噬細(xì)胞、樹突狀 細(xì)胞和某些基質(zhì)細(xì)胞產(chǎn)生，在 B 細(xì)胞的存活、增殖和成熟中起關(guān)鍵作用。 BAFF 通過與受體結(jié)合，調(diào)節(jié) B 細(xì)胞的發(fā)育和功能，其過度表達與自身免疫性疾病 （如系統(tǒng)性紅斑狼瘡、類風(fēng)濕性關(guān)節(jié)炎）密切相關(guān)。BAFF 有三個主要受體：BAFF-R （BR3）、TACI 和 BCMA。BAFF 與 BAFF-R 結(jié)合后，激活 NF-κB 和 PI3K/AKT 通路，促 進 B 細(xì)胞的存活和增殖；TACI 介導(dǎo) BAFF 和 APRIL（另一種 TNF 家族成員）的信號傳導(dǎo)， 調(diào)節(jié) B 細(xì)胞的活化和抗體產(chǎn)生；BCMA 主要在漿細(xì)胞中表達，介導(dǎo) BAFF 和 APRIL 的信 號傳導(dǎo)，支持漿細(xì)胞的存活。BAFF 的過度表達與系統(tǒng)性紅斑狼瘡（SLE）、類風(fēng)濕性關(guān)節(jié) 炎（RA）、干燥綜合等自身免疫性疾病密切相關(guān)。 | |
