| I. Background | |
| 白細(xì)胞介素 7（IL-7）是適應(yīng)性免疫系統(tǒng)所必需的細(xì)胞因子，對(duì) B 細(xì)胞發(fā)育以及記憶 和初始 T 細(xì)胞的增殖和存活以及胸腺中的 T 細(xì)胞發(fā)育至關(guān)重要。IL-7 在許多組織中廣泛表 達(dá)，包括骨髓、胸腺、淋巴結(jié)和脾等淋巴器官，以及皮膚、肺、腸和肝臟等非淋巴部位。 IL-7 主要通過激活 IL-7 受體（IL-7R）來發(fā)揮其生物學(xué)功能。IL7R 是一種異二聚體， 由 IL-7、IL-7、IL-4、IL-9、IL-15 和 IL-21 等多種細(xì)胞因子共有的 IL-7R α鏈（CD127）和 共同γ鏈（CD132， IL-2Rγ）組成。IL-7 和 IL-7Rα主要通過激活 JAK、STAT 以及 PI3K- AKT 介導(dǎo)的信號(hào)通路來促進(jìn)細(xì)胞存活并抑制細(xì)胞凋亡。IL-7 具有很強(qiáng)的免疫調(diào)節(jié)作用，可 以直接或間接作用于腫瘤細(xì)胞，通過增強(qiáng)腫瘤或過繼免疫來發(fā)揮抗腫瘤作用。然而， IL-7 也通過激活下游 JAK/STAT 和 PI3K-AKT 通路具有潛在的促腫瘤作用。 | |
