| I. Background | |
| 跨膜激活劑和 CAML 相互作用子（TACI）是腫瘤壞死因子受體（TNFR）超家族的成 員，也被稱(chēng)為 TNFRSF13B。TACI 是免疫反應(yīng)的調(diào)節(jié)因子，可抑制 B 細(xì)胞擴(kuò)增，促進(jìn)漿細(xì) 胞分化和存活。 TACI 配體是 BAFF 和 APRIL。BAFF 是屬于 TNF 配體超家族的 II 型跨膜蛋白，以膜 結(jié)合形式和可溶形式的三聚體兩種形式存在。APRIL 屬于配體 TNF 超家族，是 II 型跨膜蛋 白，參與晚期 B 細(xì)胞分化、抗體分泌漿細(xì)胞的產(chǎn)生和維持以及 IgA 類(lèi)轉(zhuǎn)換。髓系細(xì)胞（包 括巨噬細(xì)胞、樹(shù)突狀細(xì)胞和血液?jiǎn)魏思?xì)胞）以及多形核細(xì)胞（如中性粒細(xì)胞、嗜酸性粒細(xì) 胞和黏膜上皮細(xì)胞）負(fù)責(zé) APRIL 的產(chǎn)生。APRIL 對(duì) BCMA 表現(xiàn)出強(qiáng)結(jié)合親和力，對(duì) TACI 具有中等親和力，而 BAFF 與 BCMA 的結(jié)合較弱，與 TACI 具有較強(qiáng)的親和力。TACI 與 APRIL 和硫酸乙酰肝素蛋白聚糖 （HSPG） 相互作用，促進(jìn) TACI 高效信號(hào)傳導(dǎo)以發(fā)揮其 生理效應(yīng)。 | |
